The purpose of Volume 12 is to provide a comprehensive overview of the field of chemical and radiation carcinogenesis with particular emphasis on molecular mechanisms. The actions of various chemical carcinogens as well as ultraviolet and ionizing radiation are discussed in the context of the multistage process of both experimental and human carcinogenesis. Background information concerning the target genes for the genetic and epigenetic modulation by carcinogens are covered. These target genes include proto-oncogenes, tumor suppressor genes, and effector genes. Various classes of carcinogens including tumor initiating, promoting, and progressing agents are discussed in terms of how they bring about critical alterations in the target genes. The functional role of these gene alterations are covered in the context of the various phenotypic changes associated with each stage of carcinogenesis. Finally, strategies for intervening with the multistep process of carcinogenesis through chemoprevention are discussed. Wherever possible the relevancy of more basic findings with experimental model systems to human carcinogenesis is emphasized.
