In human-computer interaction this kind of use is more often described as intuitive. We define familiarity as the readiness to engage with technology which arises from being repeatedly exposed to it-often from birth. This exposure involves the frequent use of it and seeing people all around us using it every day. Digital technology has become as common a feature of our everyday lives as the motor car, TV, credit card, cutlery, or a dozen other things which we also use without conscious deliberation. We will argue that we cope with digital technology in the same way as we do these other technologies by means of this everyday familiarity. But this is only half of the story. We also regularly support or scaffold our use of technology. These scaffolding activities are described as epistemic actions which we adopt to make it easier for us to accomplish our goals. With digital technology these epistemic actions include appropriating it to more closer meet our needs. Ca2+ is a major regulator of VSM contraction and is strictly regulated by an intricate system of Ca2+ mobilization and Ca2+ homeostatic mechanisms. The interaction of a physiological agonist with its plasma membrane receptor stimulates the hydrolysis of membrane phospholipids and increases the generation of inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG). IP3 stimulates Ca2+ release from the intracellular stores in the sarcoplasmic reticulum. Agonists also stimulate Ca2+ influx from the extracellular space via voltage-gated, receptor-operated, and store-operated channels. Ca2+ homeostatic mechanisms tend to decrease the intracellular free Ca2+ concentration ([Ca2+]i) by activating Ca2+ extrusion via the plasmalemmal Ca2+ pump and the Na+/Ca2+ exchanger and the uptake of excess Ca2+ by the sarcoplasmic reticulum and possibly the mitochondria. A threshold increase in [Ca2+]i activates Ca2+-dependent myosin light chain (MLC) phosphorylation, stimulates actin-myosin interaction, and initiates VSM contraction. The agonist-induced maintained increase in DAG also activates specific protein kinase C (PKC) isoforms, which in turn cause phosphorylation of cytoplasmic substrates that increase the contractile myofilaments force sensitivity to Ca2+ and thereby enhance VSM contraction. Agonists could also activate Rho kinase (ROCK), leading to inhibition of MLC phosphatase and further enhancement of the myofilaments force sensitivity to Ca2+. The combined increases in [Ca2+]i, PKC and ROCK activity cause significant vasoconstriction and could also stimulate VSM hypertrophy and hyperplasia. These enzymes are involved in the maturation of many precursor proteins involved in the process of tumorigenesis and metastasis. Since their discovery, PCs were suggested as potential targets for anti-cancer therapy, and their activity was found to directly affect tumor cell proliferation, migration invasion, and the malignant phenotypes of tumor cells. Here, we discuss a number of previous and recent findings on the PCs features, their implication in the regulation of multiple cellular functions that impact on the invasive/metastatic potential of cancer cells, and their clinical relevance in cancer patients. Among the substrates of the proprotein convertases, various growth factors, their receptors, adhesion molecules, and proteases were identified. The PCs are inhibited by endogenous and exogenous inhibitors. To date, only pro7B2, a specific chaperone of PC2, and the granine-like precursor of neuroendocrine protein proSAAS, a selective ligand of PC1, have been identified as endogenous inhibitors of the PCs found in the regulated pathway. However, only PCs prosegments, several bioengineered inhibitors, peptides, and non-peptide compounds were found to inhibit the activity of the PCs found in the secretory pathway. The mucosal defense system is stratified into pre-epithelial (alkaline mucus), epithelial (dynamic epithelial lining), and post-epithelial (microcirculation) components. The mucus lining the epithelial surface presents a diffusional barrier to ingested material (e.g., lipids) and also serves as an unstirred layer in which a pH gradient can be established to prevent acid-induced injury. The epithelial lining prevents entrance of any toxic material to the interstitium and, should it be damaged, it is rapidly resealed by migration of adjacent viable epithelial cells to cover the defect. Any acid or other material that has entered the interstitium is washed out by an intense neurogenic hyperemia. In general, the mucosal defense system is quite effective and any adverse gastrointestinal effects associated with the normal course of nutrient assimilation are minimal. However, there are two situations in which the mucosal defense system is known to be ineffective and result in gastric mucosal injury: inadvertent ingestion of H. pylori. or intentional ingestion of NSAIDs. In this book, we review various approaches to face detection developed in the past decade, with more emphasis on boosting-based learning algorithms. We then present a series of algorithms that are empowered by the statistical view of boosting and the concept of multiple instance learning. We start by describing a boosting learning framework that is capable to handle billions of training examples. It differs from traditional bootstrapping schemes in that no intermediate thresholds need to be set during training, yet the total number of negative examples used for feature selection remains constant and focused (on the poor performing ones). A multiple instance pruning scheme is then adopted to set the intermediate thresholds after boosting learning. This algorithm generates detectors that are both fast and accurate. We then present two multiple instance learning schemes for face detection, multiple instance learning boosting (MILBoost) and winner-take-all multiple category boosting (WTA-McBoost). MILBoost addresses the uncertainty in accurately pinpointing the location of the object being detected, while WTA-McBoost addresses the uncertainty in determining the most appropriate subcategory label for multiview object detection. Both schemes can resolve the ambiguity of the labeling process and reduce outliers during training, which leads to improved detector performances. In many applications, a detector trained with generic data sets may not perform optimally in a new environment. We propose detection adaption, which is a promising solution for this problem. We present an adaptation scheme based on the Taylor expansion of the boosting learning objective function, and we propose to store the second order statistics of the generic training data for future adaptation. We show that with a small amount of labeled data in the new environment, the detector's performance can be greatly improved. We also present two interesting applications where boosting learning was applied successfully. The first application is face verification for filtering and ranking image/video search results on celebrities. We present boosted multi-task learning (MTL), yet another boosting learning algorithm that extends MILBoost with a graphical model. Since the available number of training images for each celebrity may be limited, learning individual classifiers for each person may cause overfitting. MTL jointly learns classifiers for multiple people by sharing a few boosting classifiers in order to avoid overfitting. The second application addresses the need of speaker detection in conference rooms. The goal is to find who is speaking, given a microphone array and a panoramic video of the room. We show that by combining audio and visual features in a boosting framework, we can determine the speaker's position very accurately. The motivation for developing this synthesis lecture was to provide a tutorial on queuing and trunking, with extensions to networks of queues, suitable for supplementing courses in communications, stochastic processes, and networking. An essential component of this lecture is MATLAB-based demonstrations and exercises, which can be easily modified to enable the student to observe and evaluate the impact of changing parameters, arrival and departure statistics, queuing disciplines, the number of servers, and other important aspects of the underlying system model. Much of the work in this lecture is based on Poisson statistics, since Poisson models are useful due to the fact that Poisson models are analytically tractable and provide a useful approximation for many applications. We recognize that the validity of Poisson statistics is questionable for a number of networking applications and therefore we briefly discuss self-similar models and the Hurst parameter, long-term dependent models, the Pareto distribution, and other related topics. Appropriate references are given for continued study on these topics. The initial chapters of this book consider individual queues in isolation. The systems studied consist of an arrival process, a single queue with a particular queuing discipline, and one or more servers. While this allows us to study the basic concepts of queuing and trunking, modern data networks consist of many queues that interact in complex ways.
