Bien que chaque partie puisse être lue séparément, le roman a été conçu comme un tout dont l'unité n'apparaît qu'à la lecture de l'ensemble. De l'une à l'autre, les thèmes sont repris, entrelacés, rendus méconnaissables ; les questions laissées en suspens dans une partie trouvent parfois mais pas toujours des réponses dans une suivante. Le sens des questions n'apparaît que lorsque une réponse est donnée, et le sens de ces réponses n'est saisi que lorsque la question est posée. Car, si la succession des parties n'est pas arbitraire, si la lecture linéaire est possible, 2666 peut aussi se lire comme un ensemble de récits, en partie simultanés, d'une sorte de Divine Comédie dont les cercles - plutôt infernaux que paradisiaques - seraient inextricablement mêlés.L'ouvrage commence avec La partie des critiques. Un écrivain invisible et génial, Benno von Archimboldi, entraîne dans sa recherche les critiques vers Santa Teresa. Cette ville du nord du Mexique où des femmes sont tuées par un meurtrier demeuré impuni devient peu à peu le centre du texte, comme un reflet de l'horreur du monde.S'ouvre ensuite La partie d'Amalfitano, un personnage mélancolique qui apparaît à la fin de la précédente partie et qui réside à Santa Teresa depuis peu. Une atmosphère cauchemardesque se propage, accentuée par la crainte de cet homme face à la disparition de sa fille qui pourrait être une nouvelle victime du criminel.La jeune fille réapparaît dans La partie de Fate : un journaliste nord-américain, qui se retrouve à Santa Teresa par hasard, devine plus qu'il ne voit l'horreur et la mort qui le frôlent. Suffisamment pourtant pour qu'il prenne la fuite, accompagné par la fille d'Amalfitano.La partie des crimes plonge de plain pied dans l'horreur : le texte est scandé par la description des femmes mortes, ordonnée selon les dates de découverte de leurs cadavres. Le récit entremêle les péripéties de plusieurs personnages, parmi lesquelles celles d'un inspecteur, d'un jeune agent de police et d'un journaliste, tous, d'une manière ou d'une autre, liés aux assassinats et aux tentatives de résolution. Mais évidemment, il n'y aura pas de résolution.Dans la cinquième partie, Archimboldi refait surface. C'est sa biographie que nous lisons : son enfance en Allemagne après la 1ère guerre mondiale, sa participation à la 2ème guerre mondiale, sa formation d'écrivain et les raisons de sa présence à Santa Teresa.L'entreprise de Bolaño est ambitieuse.
