However, tumor blood vessels are abnormal, bothmorphologically and functionally, and display characteristics thatdistinguish them from normal vasculature. It is these inherentdifferences between blood vessels associated with tumors and thoseassociated with normal tissues that provide a variety of uniquetargets for the design of novel therapeutics and treatmentstrategies highly selective for the cancer.

Vascular-disrupting strategies aim to cause a rapid andcatastrophic shutdown in the established vessel networks of solidtumors. This arrests the blood flow and induces tumor cell death asa result of oxygen and nutrient deprivation and build up of wasteproducts. Biological vascular-disrupting approaches includetargeted gene therapy, antibodies to neovascular antigens andligand-directed therapies targeting endothelial cell receptors andextracellular matrix proteins. Small molecule drug approaches havefocused primarily on flavenoids and tubulin-binding agents. Thisbook examines the fundamental bases of both these approaches. Emphasis is placed on target development, preclinical assessment,use in combination with conventional treatment regimens and thecurrent clinical status of these therapies.
