Active pharmaceutical ingredients, natural constituents, and pharmaceutically inert excipients have been widely studied as P-gp inhibitors. Three excipients ghee, badam oil and fenugreek oil were selected as excipients as there are enriched with p-gp substrates. Molecular docking studies were conducted to identify the binding energy involved in between p-gp and the components of selected excipients. The selected drugs affinity with p-gp was also estimated by molecular docking studies. The diffusion of selected drugs through the intestine membrane was also estimated. Good correlation was observed between the docking score and the estimated diffusion rate from the diffusion data. Thus, these studies conclude that the molecular docking studies are beneficial to select the suitable excipients for improving the bioavailability of reported p-gp substrates. 52 pp. Englisch.

Pharmazeutische Wirkstoffe, natürliche Bestandteile und pharmazeutisch inerte Hilfsstoffe sind als P-gp-Inhibitoren umfassend untersucht worden. Drei Hilfsstoffe - Ghee, Badamöl und Bockshornkleeöl - wurden als Hilfsstoffe ausgewählt, da sie mit p-gp-Substraten angereichert sind. Es wurden molekulare Docking-Studien durchgeführt, um die Bindungsenergie zwischen p-gp und den Komponenten der ausgewählten Hilfsstoffe zu ermitteln. Die Affinität der ausgewählten Arzneimittel zu p-gp wurde ebenfalls durch molekulare Docking-Studien geschätzt. Die Diffusion ausgewählter Arzneimittel durch die Darmmembran wurde ebenfalls geschätzt. Es wurde eine gute Korrelation zwischen der Docking-Punktzahl und der anhand der Diffusionsdaten geschätzten Diffusionsrate festgestellt. Diese Studien lassen den Schluss zu, dass die molekularen Docking-Studien für die Auswahl geeigneter Hilfsstoffe zur Verbesserung der Bioverfügbarkeit der gemeldeten p-gp-Substrate von Nutzen sind.
