Nach Abschluss einer vierjährigen Schulzeit arbeitete Demjanjuk als Traktorist in einer Kolchose. 1940 wurde er von der Roten Armee eingezogen. Im Mai 1942 geriet er bei der Schlacht von Kertsch in deutsche Kriegsgefangenschaft. Demjanjuk kam in ein Kriegsgefangenenlager bei Chelm, wo er vermutlich als sogenannter Hilfswilliger rekrutiert wurde. Er wurde im SS-Außenlager Trawniki ausgebildet und anschließend von der SS vereidigt. Seine erste Aufgabe war die Bewachung jüdischer Zwangsarbeiter in der Landwirtschaft. Kurz darauf war er vermutlich im KZ Majdanek tätig, welche Aufgabe er dort genau hatte, ist nicht geklärt. Am 27. März 1943 wurde er vermutlich in das Vernichtungslager Sobibor abkommandiert, wo er als einer von etwa 130 Hilfswilligen unter dem Kommando von 20 bis 30 Deutschen gedient haben und zur Außensicherung des Vernichtungslagers eingesetzt worden sein soll.
