In recent years it has become clear that the molecular genetic approach can illuminate many still murky areas of Developmental Biology and in particular two of its major mysteries. 1)The molecular Bases of Cellular Differentiation - When a cell is exposed to an appropriate microenvironment, it enters upon a complex, a temporally and quantitatively regulated program of phenotypic alterations that leaves it greatly different in form and function from sister cells located elsewhere within the cell assembly. These phenotypic changes can be ascribed to ordered sequences of differential gene expression. Question: What molecular events induce mediate and control such sequences? 2) The Inheritance of Differentiative Capacity- By virtue of its position within a developing Metazoan, a given cell may yield a clone whose members are restricted to development only along certain pathways of cellular differentiation. Thus, a hematocytoblast is a stem cell whose progeny are restricted to erythropoietic development. Question: Do such heritable restrictions arise from modifications of the genetic apparatus or from self maintained feedback circuits within the cytoplasm or both?
