Sa famille est très pauvre et il est élevé par sa mère Aretha Williams à Greenville en Floride. L'enfance de Ray est marquée par des traumatismes physiques et psychologiques : après la perte de son petit frère, il doit faire face à un glaucome et perd la vue à l'âge de 7 ans. Il est alors placé dans une institution spécialisée. C'est dans cette école que, 9 années durant, il apprend la composition, ainsi que la pratique de plusieurs instruments, dont la clarinette, le saxophone alto et le piano. Malgré un enseignement musical essentiellement classique, ses préférences s'orientent rapidement vers des musiques contemporaines : le gospel, le blues, le jazz et le country. C'est avec le blues qu'il connaît le succès au début des années 1960 avec des titres tels que Georgia on My Mind ou Hit the Road Jack.
