Ordained as a Lutheran presbyter, he joined CMS, arriving in India in 1814. Rhenius engaged in missionary work in Madras, establishing schools and contributing to the revision of the Tamil Bible. In 1817, he founded the Tamil Bible Association. Departing from the Anglican Church in 1830, he started his congregation. His contributions were officially recognized in 1980. Rhenius's legacy includes the Holy Trinity Cathedral in Palayamkottai and "A Grammar of the Tamil Language: With Appendix" (1859).
