Sein hauptsächliches Forschungsgebiet waren die veränderlichen Sterne. Er hat mehr als ein Viertel der heute bekannten veränderlichen Sterne entdeckt. Dazu gründete er Mitte der 1920er Jahre die Sternwarte Sonneberg und entwickelte die Langzeit-Forschungsprogramme "Sonneberger Felderplan" (Field patrol) und "Sonneberger Himmelsüberwachung" (Sky patrol) zur Beobachtung und fotografischen Überwachung und Erforschung der veränderlichen Sterne, Meteore und extragalaktischer Objekte. Unter seiner Leitung wurde die Sternwarte in Sonneberg bis Ende der 1960er Jahre zum weltweit führenden Institut auf diesem Spezialgebiet der Astronomie. Zum Gedenken gab 1970 die Internationale Astronomische Union (IAU) dem Mondkrater mit den Koordinaten "15° 12' N / 136° 54' O" den Namen Hoffmeister.
