Fischer received the Nobel Prize in Chemistry in 1902. "Berlin chemist Emil Fischer and Joseph von Mering synthesized diethyl barbituric acid or barb…itone (Veronal) in 1902. They described their discovery in "Ueber eine neue Klasse von Schlafmitteln", Therapie Gegenwart 44:97-101, 1903. The parent compound, barbituric acid, was first synthesised by Adolf von Baeyer in 1864, but barbituric acid is not itself pharmacologically active. Fischer and von Mering soon realized that their new drug was a sedative. Veronal represented a huge improvement on the medley of existing agents. It didn't taste unpleasant. It had few adverse side effects. Unlike foul-tasting potassium bromide, Veronal acted at therapeutic levels far beneath the toxic dose. Some 2,500 barbiturates were synthesized over the next century. More than fifty have been marketed as sedatives. In 1912, phenobarbital was introduced under the brand-name Luminal. From 1912 until around 1960 barbiturates were the mainstay of pharmacological treatments of anxiety and insomnia. The development of soluble barbiturates suitable for use as intravenous anaesthetics still lay in the future." This led to the seminal discovery of the hypnotic action of diethylbarbituric acid." This work introduced an entirely new synthetic drug called Veronal, a barbiturate family still of major importance in clinical medicine. [Gedeon]. Barbital (or barbitone), marketed under the brand names Veronal for the pure acid and Medinal for the sodium salt, was the first commercially available barbiturate. It was used as a sleeping aid (hypnotic) from 1903 until the mid-1950s. / "Barbital was first produced in 1882 by Max Conrad and Max Guthzeit without further identification. Twenty years later, in 1902, it was first synthesized by the German Emil Fischer and his nephew Alfred Dilthey and characterized as a sleeping pill by Josef von Mering. Barbital was brought onto the market in tablet form by Merck in 1903 as the first barbiturate under the brand name Veronal. It had not become established as an anesthetic because of its toxic side effects in higher doses, but was used as a strong sedative as part of premedication before anaesthesia. In combination with amidopyrine, it was advertised for many years as Veramon, among others, also as a remedy for menstrual cramps, sold by Schering-Kahlbaum AG. Chemie, die er durch wichtige Arbeiten über Nitrobenzolvergiftung, Diabetes, Glycogenbildung in der Leber, Abzugswege des Zuckers aus der Darmhöhle, Umwandlung von Stärke und Glycogen durch Diastase, Speichel, Pankreas und Labferment, über den Einfluss von diastalischen Fermenten auf Stärke, Dextrin und Maltose, über Zuckerausscheidung nach Phloridcin-Gebrauch, nach Zerstörung der Bauchspeicheldrüse, über die schlafmachende Wirkung des Amylenhydrats, über chlorsaures Kali, Lipanin als Ersatz des Leberthrans, Verhalten der Magensäuren, über das Schicksal des Chloralhydrats im Organismus, des Kairins im Tierkörper, über künstliche Osteomalacie, Phosphorvergiftung, Wirkung des Quecksilbers und Ferrocyankaliums, Chloride im Hundeharn, Nahrungszufuhr und Oxydationsprozesse u.a.m. förderte. "Als wichtigster und erfolgreichster Weg erwies sich indes die Untersuchung der cyclischen Carbamidderivate, deren Ursprung auf die Purinforschung EMIL… FISCHERS (1852-1919), des größten Chemikers der Jahrhundertwende, zurückgeführt werden kann. FISCHER begann seine Harnsäureshrte.ynthese, indem er Carbamid mit Malonsäure kondensierte. So stellte er 1895 zuerst Malonylharnstoff her, der später nach einem Mädchen Barbara Barbitursäure genannt wurde. Diese war noch nicht narkotisch; als er jedoch am C5 zwei Äthylgruppen substituierte, was damals (1903) nach dem Beispiel des Sulfonals naheliegend schien, gewann er die stark wirkende Diäthylbarbitursäure. Die ersten Tierversuche führte MEHRING durch, der alsbald erfreut an FISCHER telegraphierte: »Veronal ausgezeichnet, Hund frißt sofort nach Erwachen.« Das neue Schlafmittel wird nach der Stadt Verona Veronal genannt. Obwohl Veronal rasche Verbreitung fand, setzten FISCHER und MEHRING ihre Arbeit fort und stellten höhere Homologe des Veronals her. Schon 1905 veröffentlichte sie eine Mitteilung über Proponal, das doppelt so stark wirkte wie Veronal.
