This book deals with the traditionaluse of plants to treat TB-like symptoms and attemptsto establish a link between naphthoquinones and theseplants with the use of a metabolomics approach. Itfurthermore deals with the chemical synthesis of themain lead compound and some derivatives. This isfollowed by the often neglected stability studiesperformed on the main active compounds. These studiesshowed that rapid breakdown of lead compounds dooccur during activity and toxicity testing, and thatthis leads to false positives and negatives. Theactivity and toxicity of the main active compoundswere tested using three models: Vero cells, mice andMusca domestica. The last chapters deal with theinvestigation of a structure activity relationshipand propose a mechanism of action for thesenaphthoquinones. It was found that this class ofcompounds interfere with the electron transport chainby inhibiting menaquinone to bind to the cytochromecomplex. This indicates that these compounds mightbecome a new class of antibiotics.…
