Cet ouvrage, très ambitieux sur le plan philosophique, est un jalon très important dans l itinéraire de Nicolas de Cues. Il formule là pour la première fois la philosophie de l esprit, qu il développera plus avant dans des ouvrages ultérieurs tels que « Le Profane sur l esprit », le « Sur la sagesse », « La Chasse de la sagesse », etc. L ouvrage influencera d une façon profonde Jean Pic de la Mirandole (1463-1494) et à travers lui les cercles humanistes de Florence. On trouve des traces plus ou moins importantes des doctrines cusaines chez Montaigne (1533-1592), Bruno (1548-1600), Descartes (1596-1650) ou Leibniz (1646-1716). « Les Conjectures » constituent une transition intellectuelle entre le Moyen Âge et la Renaissance de première importance.
