Het was gesticht door Jacob de Vos en Henriëtte Wurfbain, rijke Amsterdammers die hun vermogen gebruikten voor bijzondere dingen. Ze steunden uiteenlopende projecten: onderwijs, barmhartigheid en armenzorg, maar toonden ook een grote passie voor met name de beeldende kunsten. Jacob de Vos is vooral bekend geworden omdat hij de grootste collectie tekeningen van oude meesters van Nederland opbouwde en met zijn vaderlandsliefde als mecenas optrad om de historieschilderkunst te bevorderen. Met een uniek schilderproject creëerde hij een 'klein Versailles' in zijn stadspaleisje aan de Herengracht. De veiling van zijn collectie vormde de directe aanleiding tot de oprichting van de Vereniging Rembrandt en met zijn tekeningen kon de collectie van het Rijksprentenkabinet in één klap worden verdubbeld. Het verdwenen hofje beschrijft de bouw- en bewoningsgeschiedenis van het hofje en tegelijk de fascinerende geschiedenis van leven en erfgoed van het echtpaar De Vos-Wurfbain, in het bijzonder van Jacobs mecenaat, collectie en verzamelmanie.
