"Neben die Auswertung der Befunde tritt in der zweiten Hälfte des vorigen (19.Jh.) Jahrhunderts die Frage nach der Ursache der Doppelbrechung tierischer und pflanzlicher Strukturen. Zwar kündet optische Anisotropie stets die Anwesenheit eines sub- oder amikroskopischen anisometrischen Feinbaues an; aber wie dieser beschaffen, darüber gingen die Meinungen anfangs sehr auseinander: EHRENBERG (1849), BRÜCKE (1858: Disdiaklastenhypothese), Hugo von MOHL (1859) und insbesondere C.v.NAEGELI (1859; 1862; 1879: Micellarhypothese) sprachen sich für Kristallinität aus, CH.ROUGET (1862) und W.HOFMEISTER (1867) für die Wirksamkeit feinster lichtbeugender Lamellierungen oder Streifungen (Depolarisationshypothese); v. EBNER (1882) aber nahm an, die an sich isotropen Teilchen der Gewebe erhielten durch Spannungen während des Wachstums nach verschiedenen Richtungen unterschiedliche Abstände (Spannungshypothese). Die durch scheinbar widersprechende Beobachtungen sehr verwickelte Sachlage klärten in den Jahren 1910 bis 1919 H.AMBRONNs Untersuchungen an Gallerten organismischer Herkunft, wie Gelatine und Celloidin. Er gelangte nicht nur zu einer Trennung von Orientierungsdoppelbrechung und photoelastischem Effekt, die bis dahin bei solchen Objekten vielfach noch zusammengeworfen wurden, sondern er konnte bei der Orientierungsdoppelbrechung zwei Anteile von ganz verschiedener Art unterscheiden, die Texturdoppelbrechung, die durch die Anisometrie eines submikroskopischen Feinbaues nach Art eines WIENERschen Mischköpers hervorgerufen wird, und die Eigendoppelbrechung, die auf amikroskopischem Feinbau der Texturelemente selbst zu beziehen ist, der bald darauf durch das Röntgen-Faserdiagramm als raumgittermäßige Ordnung gesichert wurde. Auf dieser Grundlage und unter Beachtung der neueren Einsichten in die Stereochemie der Baustoffe ließ sich die Polarisationsoptik von Zell- und Gewebestrukturen als hydrogele und lipoide Schichtsysteme befriedigend deuten und zugleich für die Analyse des Feinbaues verwenden."
