Dupérié était depuis plusieurs années un élève de HAYEM qui lui inspira cette thèse, basée en grande partie sur des recherches personnelles. Il avait déjà recueilli en 1875 la leçon de Hayem sur la "Numération des globules du sang", leçon publiée dans la Gazette hebdomadaire de médecine et de chirurgie. Dupérié étudie ici les méthodes de numération, les valeurs normales et les variations suivant l'âge, le sexe, l'alimentation. On trouve in fine un tableau synoptique. Hayem, dans ses "Recherches sur l'anatomie normale et pathologique du sang" (paru la même année), indique (p. 11) qu'il fut aidé par Dupérié dans ses numérations. Cependant l'ouvrage de Hayem dans lequel il expose précisément ces variations physiologiques est son grand traité intitulé "Du sang et de ses altérations anatomiques" qui ne paraîtra qu'en 1889. Hayem y écrit (p. 178) : "J'ai été très aidé dans cette partie de mon travail par mes élèves MM. Dupérié et Cadet qui ont consacré leur thèse inaugurale à cette étude".
