The underlyingmechanisms are poorly understood bu…t are likelyrelated to the abnormal mechanical properties ofleukemia cells. Using atomic force microscopy andmicrofluidic devices to quantify cell deformation, Ifound that leukemia cells taken from patients withleukostasis were significantly stiffer than leukemiacells taken from asymptomatic patients. These resultssuggest that increased leukemia cell stiffness may bean independent leukostasis risk factor and could beuseful for predicting leukostasis. In addition, Ifound that chemotherapy increases leukemia cellstiffness by up to 100x and therefore, may actuallyincrease leukostasis risk. However, thephosphodiesterase inhibitor pentoxifyllinesubstantially decreased leukemia cell stiffness andadhesiveness, and this drug may be a potentialtreatment for leukostasis. Overall, these studiesshow that cell mechanics are relevant in disease. Inaddition, tools that measure cell mechanics can beuseful diagnostic/drug discovery platforms.
