Nach seinem ersten Biographen Ridolfi wurde Guidobaldo II. della Rovere, der Herzog von Urbino auf ihn aufmerksam und ermöglichte einen Studienaufenthalt an seinem Hof in Pesaro. 1564-67 studierte Palma die herzogliche Sammlung. Darauf wurde er zu weiterem Studium nach Rom gesandt. 1570 nach Venedig zurückgekehrt, brauchte er bis circa 1580, um einen eigenen Stil zu entwickeln, und sich in die Eigentümlichkeiten der venezianischen Schule des Spätmanierismus einzufügen. Nach dem ersten der zwei Brände im Dogenpalast erhielt Palma 1577 einen ersten Auftrag im Kontext der Wiederherstellung der Palasträume. 1583 wurde Palma zur Ausmalung des Oratoriums im Hospiz des Ordens der Crociferi engagiert. In der Folge malte er noch einige Gemälde für die Kirche (Santa Maria Assunta). Neben öffentlichen und kirchlichen Aufträgen, wurde er auch für private Mäzene tätig. Nach Tintorettos Tod im Jahre 1594 galt er bereits als der führende Maler Venedigs. Seine kunstgeschichtliche Bedeutung liegt vor allem in seiner Rolle als Wegbereiter des Frühbarock in Venedig.
