Es ist in einem großen Blutgefäß des Egels lokalisiert und ständig von Blut umspült. Blutegelblut enthält verschiedene organische Säurereste, welche alle Intermediärprodukte des Citratzyklus' sind. Der prominenteste organische Säurerest ist Malat.Diese Arbeit untersucht die Bedeutung dieser Blutbestandteile für die Funktion des Zentralnervensystems des Blutegels. Des Weiteren wird eine Malat-haltige Ringerlösung entwickelt und getestet, die geeignet ist, die Lebensdauer isolierter Teile des Blutegel-ZNS von einem Tag auf eine Woche zu verlängern, ohne auf Zellkulturmedien zurückzugreifen.Es wurde die Wirkung von Glucose, Pyruvat und Malat auf Funktion und Lebensdauer von Segmentalganglien des Zentralnervensystems des Blutegels untersucht. Hierzu wurden isolierte Segmentalganglien von Hirudo verbana in Lösungen mit 10 mM Glucose, 5 mM Pyruvat, 15 mM Malat, bzw. einer Lösung, die frei von diesen Substanzen war, aufbewahrt. Zu definierten Zeiten wurden die elektrophysiologischen Parameter zweier verschiedener Neuronen, sowie von Gliazellen mittels intrazellulärer Ableitung und Applikation von Neurotransmittern bestimmt. Darüber hinaus wurde die mechanische Konsistenz und die lichtmikroskopisch erfassbare Morphologie der Segmentalganglien dokumentiert.-----------------------The central nervous system of the medicinal leech Hirudo medicinalis is a well-studied neurophysiological model system. In situ, it is located inside the leech's major blood vessel and constantly surrounded by blood flow (Sawyer 1986). Leech blood contains various inorganic cations and carboxylate anions, with malate being the most prominent component (Nieczaj & Zerbst-Boroffka 1993).My work discusses the significance of these carboxylate anions for the function of the leech central nervous system. Furthermore, I propose and test the use of a malate-supplemented Ringer's solution to nourish isolated parts of the leech CNS and prolong their lifetime from one day to a week without the use of cell culture media.
