Des dispersions solides de bosentan ont ete preparees par fusion a l'aide de deux excipients hydrophiles fusibles selectionnes, le gelucire 50/13 et le poloxamere 188. Le Sylysia 350 a ete utilise comme adsorbant. Les etudes de solubilite ont montre une augmentation de 8 et 10 fois la solubilite pour les dispersions solides a base de gelucire 50/13 et de poloxamere 188, respectivement. Les valeurs d'energie libre de Gibbs DGtr0 etaient toutes negatives pour la gelucire 50/13 et le poloxamere 188, indiquant la nature spontanee de la solubilisation. Les dispersions solides presentant la solubilite la plus elevee ont ete comprimees en comprimes a liberation immediate a l'aide de glycolate de sodium et d'amidon comme superdesintegrant. Les etudes de dissolution in vitro ont montre une dissolution du medicament superieure a 90 % en 1 heure. Le gelucire 50/13 et le poloxamere 188 jouent un role important dans l'amelioration de la solubilite et de la dissolution du medicament.L'adsorbant Sylysia 350 peut etre utilise pour conferer une bonne fluidite et compressibilite aux dispersions solides. Solid dispersions of bosentan were prepared by fusion method by using two selected hydrophilic meltable carriers vis-à-vis gelucire 50/13 and poloxamer 188. Sylysia 350 was used as an adsorbent. Solubility studies showed 8 and 10 fold increase in solubility for gelucire 50/13 and poloxamer 188 based solid dispersions respectively. The Gibbs free energy DeltaGtr° values were all negative for gelucire 50/13 and poloxamer 188 indicating spontaneous nature of solubilisation. Solid dispersions exhibiting highest solubility were compressed into immediate release tablets by using sodium starch glycolate as superdisintegrant. In vitro dissolution studies, exhibited more than 90 % drug dissolution in 1 h. Gelucire 50/13 and poloxamer 188 plays a significant role in enhancement of drug solubility and dissolution.The adsorbent, sylysia 350 may be used to impart good flow and compressibility to solid dispersions. Among the two carriers, poloxamer 188 exhibited better solubility and dissolution enhancement potential. 72 pp. Englisch.
