Rost studierte in Greifswald, Leipzig und Heidelberg, wurde 1896 promoviert (Ueber die Ausscheidung des Coffein und Theobromin im Harn') und war Assistent am Pharmakologischen Institut in Marburg/Lahn. Seit 1898 war er Vorstand des Physiologisch-Pharmakologischen Instituts im Reichsgesundheitsamt in Berlin, habilitierte sich 1901 für Pharmakologie und erhielt 1912 eine a.o. Professur. Er arbeitete vor allem auf den Gebieten Pharmakologie, Toxikologie und Ernährungsphysiologie. Er veröffentlichte u.a. ein Handbuch der allgemeinen und speziellen Arzneiverordnungslehre, Pharmacopea universalis (1929).
