Cet ouvrage comporte un exposé fidèle des premiers travaux auxquels s'est livré le Comité, avec les principaux résultats obtenus, ayant joué un rôle important dans la diffusion de la vaccine. Il est divisé en trois parties. La première partie (pp. 1-63) comporte de précieuses données historiques : travaux de Jenner, attention donnée à la vaccine en France, formation du comité, exposé des essais faits en France, relations entretenues par le comité avec les médecins des départements et avec les médecins et les gouvernements étrangers. La deuxième partie (pp. 63-174) comporte l'examen des effets de la vaccine, notamment de sa bénignité et de sa faculté préservative (exposé des nombreuses contre-épreuves). Dans la troisième partie (pp. 174-416) le comité examine les objections faites contre la vaccine. On trouve in fine la conclusion du rapport (réalité reconnue des avantages attribués à la vaccine ; services éminents qu'elle doit rendre à la société - moyen de préservation de la petite vérole, préférable à l'inoculation ordinaire, et certitude acquise de pouvoir, par elle, anéantir la petite vérole - ) ; indications des moyens propres à rendre générale l'inoculation de la vaccine. Les 11 pages insérées entre les pages 416 et 417 correspondent à un rapport du comité central de vaccine sur les "allégations" avancées par Alphonse Leroy qui, après avoir fait les éloges de l'inoculation de la vaccine, estimait maintenant qu'elle produisait des accidents graves. Le très long rapport du 11 mars 1803, sur la vaccine, du Comité central de vaccine, ainsi qu'un autre rédigé par l'Institut National, furent présentés à Napoléon Bonaparte, premier consul, qui chargea Chaptal, ministre de l'Intérieur et médecin, d'organiser une grande enquête sur la vaccination. Le 21 octobre 1803, Jenner fut nommé membre correspondant associé de l'Ecole de médecine de Paris.".
