DOS requires the development of new methodologies that generate scaffold diversity in addition to appendage and stereochemical dive…rsity. MCRs have been demonstrated to be extremely useful for DOS, since these complexity generating reactions can easily be combined with several follow-up cyclization reactions resulting in the rapid synthesis of diverse (heterocyclic) scaffolds. Furthermore, novel MCR design strategies have emerged as important tools to generate scaffold diversity. Four different approaches (SRR, MRS, CBD and MCR2) have been applied, which all led to the development of new MCRs and higher order MCRs, thereby addressing both molecular diversity and complexity. Present work is mainly based on literature review done for the research work. From the first ever reported multicomponent reaction (by Strecker in 1850) to modern multicomponent synthesis is included in the work. With proper citation a brief history of work done in area of multicomponent reaction is presented. Novel multi-component strategies towards scaffold diversity are also described. 60 pp. Englisch.

Die vorliegende Arbeit stützt sich hauptsächlich auf die für die Forschungsarbeit durchgeführte Literaturrecherche. Von der ersten jemals berichteten Mehrkomponentenreaktion (von Strecker im Jahr 1850) bis hin zur modernen Mehrkomponentensynthese wird in dieser Arbeit alles behandelt. Mit angemessenen Zitaten wird eine kurze Geschichte der Arbeiten auf dem Gebiet der Mehrkomponentenreaktion präsentiert. Inoltre, le nuove strategie di progettazione MCR sono emerse come strumenti importanti per generare la diversità degli scaffold. Sono stati applicati quattro diversi approcci (SRR, MRS, CBD e MCR2), che hanno tutti portato allo sviluppo di nuovi MCR e di MCR di ordine superiore, affrontando così sia la diversità che la complessità molecolare. Il presente lavoro si basa principalmente sulla revisione della letteratura effettuata per il lavoro di ricerca. Il lavoro include la prima reazione multicomponente mai riportata (da Strecker nel 1850) fino alla moderna sintesi multicomponente.
