Expuso los resultados de sus investigaciones en Monumentos del arte antiguo mexicano; Alfabetos aztecas; Nomenclatura geográfica, etimológica y jeroglífica de México, etc. Nacido en el seno de una humilde familia radicada en Atotonilco el Grande, municipio del hoy estado de Hidalgo, Antonio Peñafiel Barranco es, sin duda alguna, uno de los más grandes polígrafos de México y tal vez el más ilustre hidalguense de finales del siglo 19 y principios del 20. En los esfuerzos para integrar su biografía se ha incurrido en diversos desatinos, el primero derivado de su fecha de nacimiento, pues existe quien asegura que éste ocurrió en 1839, sin embargo, el descubrimiento de su testamento permite asegurar que la fecha correcta es el 1º de enero de 1830. Por otra parte, se sabe que sus primeros estudios se realizaron en la escuela "Amiga", de Atotonilco el Grande, en el hoy estado de Hidalgo, de donde pasó a la Ciudad de México, a fin de matricularse en la Escuela Nacional de Medicina. Desde luego resulta falso e incongruente que Peñafiel hubiese estudiado en el Instituto Literario del Estado de Hidalgo (hoy Universidad Autónoma de Hidalgo), dado que dicho plantel se funda el 3 de marzo de 1869, cuando el atotonilquense contaba ya con 39 años. Asimismo, debe desecharse que interrumpiera su carrera para combatir la intervención en 1862, aunque desde luego sí participa en las acciones militares, al lado del General Ignacio Zaragoza, en su carácter de médico militar, dado que se desempeñaba como profesor en el Hospital Militar de San Lucas, en la Ciudad de México. Concluido el Imperio de Maximiliano y restaurada la república, radicó por espacio de nueve años en la ciudad de Pachuca, donde se desempeñaría como profesor del Instituto Literario, fundado en el mismo año en que se creó el Estado de Hidalgo; entre 1873 y 1875, durante el gobierno de Lerdo de Tejada, fue electo como diputado federal por su estado natal. Su espíritu inquieto le llevó a realizar diversas tareas de carácter histórico, pero ante todo se le considera como uno de los primeros y más completos estadígrafos de México, lo que le llevó a formar parte del Instituto Internacional de Estadística, de la Sociedad Mexicana de Geografía y Estadística, de las también Academias: Científica Antonio Alzate, de Economía Política de París y de Filología Americana, ello, amén de fundar la Sociedad de Historia Natural. Como hombre de ciencia, promovió la edición de numerosas obras relativas a la historia antigua de México y fue autor de las obras: "Monumentos del arte mexicano", "Memoria sobre las aguas potables de la ciudad de México", "Nomenclatura geográfico, etimológica y jeroglífica, nombres geográficos de México", "Teotihuacan: estudio histórico y arqueológico", "Indumentaria antigua mexicana", "Cerámica mexicana y loza de Talavera de Puebla", "Época colonial y moderna", "Ciudades coloniales y capitales de la República Mexicana", entre las más conocidas. El presidente Porfirio Díaz le nombró director general de Estadística y en 1895 dirigió el Primer Censo General de la República, levantado en un solo día, el 20 de octubre, cuya publicación cuidadosamente revisada, logró satisfacer la aspiración tantas veces frustrada en al menos las dos décadas anteriores. Peñafiel permaneció al menos hasta mediados de la siguiente década en la Dirección de Estadística, periodo en el que se realizaron el segundo y tercer Censos Generales de Población y vivió para ver la ejecución del cuarto conteo censal en 1921. En 1919 visitó la ciudad de Pachuca con motivo de conmemorarse el cincuentenario de la fundación del Instituto Literario, ocasión en la que entregó, además de sus obras impresas, una magnífica colección de dibujos sobre diversos conventos construidos en tierras hidalgu.
