Die Demenz vom Alzheimer-Typ weist weltweit steigende Prävalenzen und Inzidenzen auf und wird in der Zukunft in noch stärkerem Umfang als bisher eine massive medizinische und soziale Herausforderung darstellen. Dieser Problematik steht eine bisher unbefriedigende Therapie der Erkrankung gegenüber, da sich Pharmaka aus der Klasse der Nootropika als weitgehend unwirksam erwiesen haben. Hinsichtlich der mnestischen Stà rungen, die bei der Alzheimer'schen Demenz auftreten, wird der cholinergen Degeneration im basalen Vorderhirn eine wesentliche Bedeutung beigemessen. Aufgrund dessen ist es ein Ziel der pharmakologischen Therapie, die cholinerge Transmission des basalen Vorderhirns zu verbessern. Eine derartige Aktivierung der cholinergen Neurone ist eventuell mit Substanzen aus der Klasse der Ã-Carboline mà glich. Dieser hypothetische therapeutische Ansatz wird in der Arbeit von T. Steckler dargestellt. Des weiteren werden Ergebnisse dreier verhaltenspharmakologischer Experimente diskutiert: Ratten mit (1) Läsionen des cholinergen basalen Vorderhirns, (2) mit Läsionen neocorticaler cholinerger Projektionsareale, oder (3) mit systemischer anticholinerger Scopolaminbehandlung explorierten ein radiales Tunnellabyrinth.
