Er stammte aus Chalkedon am Bosporus, trat in jungen Jahren in Platons Akademie ein und begleitete seinen Lehrer auf Reisen nach Sizilien; nach Platons Tod wurde er zunächst Stellvertreter Speusippos', dem er 339/338 v. Chr. als Leiter der Akademie nachfolgte. Xenokrates entwickelte eine metaphysische Lehre, in der Zahlen und mathematische Strukturen zentrale Rolle spielen, und prägte die akademische Ethik, indem er Tugend als Form vernünftiger Seelenordnung fasste; seine Schriften sind verloren, aber über antike Berichte, u. a. bei Diogenes Laertios, fragmentarisch überliefert. Medizinhistorisch ist Xenokrates vor allem indirekt von Bedeutung, da seine Lehre und die akademische Tradition über Galen und andere spätere Autoren auf medizinische Anthropologie und Seelenlehre wirkten; direkte medizinische Schriften von ihm sind nicht überliefert.
