Metformin hydrochloride is an oral antidiabetic biguinide agent, used in the management of non-insulin dependent (type-2) diabetes Mellitus. Orodispersible tablets of Metformin Hcl were prepared by direct compression method. About nine formulations for the present study were carried out based on combination of two superdisintegrants. Croscarmellose sodium, crosspovidone and sodium starch glycolate were used as superdisintegrant. The prepared batches of tablets were evaluated for weight variation, hardness, friability, wetting time, invitro dispersion time, drug content and invitro dissolution studies. The formulation containing combination of croscarmellose sodium and sodium starch glycolate showed rapid invitro dispersion time as compared to other formulations. The optimized formulation dispersed in 28 seconds. It also showed a higher water absorption ratio and 98.37% of drug is released within 30 minutes. 88 pp. Englisch.…

A dual component tablet made of a sustained release tableted core and an immediate release tableted coat was prepared by direct compression. Both the core and the coat contained a model drug (Diclofenac Sodium). The sustained release effect was achieved with a polymer (Microcrystalline cellulose) to modulate the release of the drug. The in vitro drug release profile from these tablets showed the desired biphasic release behavior: the Diclofenac Sodium contained in the fast releasing component was dissolved within 2 minutes, whereas the drug in the core tablet was released at different times (=16 or 924 hours), depending on the composition of the matrix tablet. Un comprime a double composant, constitue d'un noyau a liberation prolongee et d'un enrobage a liberation immediate, a ete prepare par compression directe. Le noyau et l'enrobage contenaient tous deux un principe actif modele (le diclofenac sodique). L'effet de liberation prolongee a ete obtenu grace a un polymere (cellulose microcristalline) permettant de moduler la liberation du medicament. Le profil de liberation in vitro de ces comprimes a montre le comportement de liberation biphasique souhaite le diclofenac sodique contenu dans le composant a liberation rapide s'est dissous en moins de 2 minutes, tandis que le medicament present dans le noyau du comprime a ete libere a des moments differents (16 ou 924 heures), en fonction de la composition de la matrice du comprime.
