Dem wohl bestuntersuchten PPAP Hyperforin ¿ Hauptinhaltsstoff der Arzneipflanze Echtes Johanniskraut (Hypericum perforatum L.) ¿ werden aus aktueller wissenschaftlicher Sicht eine zentrale Rolle an der antidepressiven Wirksamkeit von Johanniskraut-Extrakten sowie weitere vielversprechende pharmakologische Eigenschaften zugeschrieben. Da bei steigender Zahl und Prognose psychischer Erkrankungen der Bedarf an Johanniskraut und im Speziellen an Hyperforin nicht durch Kultivierung oder aufwändige, bisher unökonomische Vollsynthesen gedeckt werden kann, erweckt eine mögliche biotechnologische Produktion großes Interesse. Voraussetzung dafür ist die Entdeckung und Nutzbarmachung von beteiligten Enzymen des weitestgehend auf Vorhersagen beruhenden Hyperforin-Biosyntheseweges. Die vorliegende Dissertation befasst sich mit der Suche, Identifizierung, molekularbiologischen Untersuchung und funktionellen Analyse involvierter aromatischer Sekundärstoffwechsel-Prenyltransferasen aus H. perforatum. Das Wissen um ihre genetische Information und die enzymatischen Funktionen zugehöriger Genprodukte bildet die Grundlage für die zukünftige Implementierung einer möglichen ¿grünen Synthese¿ von Hyperforin und verwandten bioaktiven Naturstoffen in homologen bzw. adäquaten Produktionssystemen.
