Il più alto grado di direzione del commercio del cibo a Roma e negli Stati Pontifici (Prefetto dell'Annona) viene affidato a Andrea Spinola. "E' solo durante il pontificato di Gregorio XIII (1572-1585) che con la formalizzazione della Prefettura dell'Annona 9 si ebbe anche la costruzione del primo granaio pubblico della capitale. Infatti, tra i principali attributi del nuovo Prefetto, monsignor Andrea Spinola, figurava ora anche l'esclusiva per l'acquisto di grano in tutto lo Stato al preciso scopo di dotare la città di una scorta alimentare costante." "La prosperità familiare consentì al M. di intraprendere una promettente carriera ecclesiastica con l'acquisto delle cariche venali di abbreviatore apostolico (1560) e di protonotario (1567). In questa veste assunse anche alcune funzioni di un certo rilievo amministrativo, come quella di presidente delle Strade e presidente delle Carceri. Infine nel 1578, durante il pontificato di Gregorio XIII, acquistò per 60.000 scudi l'ufficio di auditor Camerae, che lo pose al vertice dell'amministrazione della giustizia a Roma e aprì la strada a più alti onori."
